Peptide reference standards: assigned values and traceability

Select peptide reference standards by assigned value, uncertainty and intended use. Review NIST examples, working standards and the supplier records to request.

Choose a peptide reference standard for the measurement it will support. The record should identify the material, the assigned property value, its uncertainty and its intended use. A catalogue purity label alone leaves those questions open.

Define the measurement before selecting the standard

A laboratory may need to confirm an expected molecular identity, compare a chromatographic impurity profile or assign a quantitative result. Those jobs place different demands on a reference material. A sample that helps a laboratory recognise a peak can still lack the value assignment needed for a quantitative assay.

Write the intended measurement into the enquiry. Include the exact peptide form and the result the laboratory will report. For a quantitative result, specify whether the basis is the material as supplied, a dry basis or another stated basis. Keep that basis with the result when records move between procurement, the supplier and the receiving laboratory.

Reference material, certified material and working standard

NIST separates Standard Reference Materials, Reference Materials and Research Grade Test Materials. The categories carry different levels of characterisation and value assignment. Its public comparison explains that certified values and their uncertainties belong to the stated properties of an SRM. That terminology should not be inferred from the phrase “research grade” on a supplier label. [1]

A working standard is a material a laboratory qualifies for a defined task. Its usefulness depends on the qualification record, the reference used and the limits of that task. Ask who assigned its value and which document connects it to the reference. A renamed catalogue vial does not create that record.

What a published NIST peptide certificate actually reports

The NIST SRM 998 certificate for Angiotensin I, issued on 19 May 2021, reports a certified value of 99.9 ± 0.1 weight percent. It also states that reported uncertainties use one standard error. This is a specific value with a stated basis and uncertainty convention, rather than a standalone percentage. [2]

The certificate is an external example of how a reference is documented. It is not a result for a True Research Peptides Labs product. When purchasing any reference, check the current certificate, the supplied unit and its validity conditions. SRM 998 ties certification validity to five years from shipping under its stated conditions; the issue date alone cannot establish the expiry date of a particular shipped unit.

Keep uncertainty attached to the assigned value

When a supplier gives an uncertainty, ask how it is expressed. A standard uncertainty, an expanded uncertainty and a confidence interval are different descriptions. Copying a number into a procurement spreadsheet while dropping its convention can make two records appear comparable when they are not.

Separate metrological traceability from lot traceability

The JCGM International Vocabulary of Metrology connects metrological traceability to a documented chain of calibrations, with uncertainty contributions. It also distinguishes that concept from tracing the history of a sample or document. [3] A batch number can establish which vial was received. It cannot, by itself, establish the reference chain for a reported concentration.

Record Question it answers What to check
Material identity Which chemical form is supplied? Peptide identity, modifications and relevant counter-ion information.
Assigned value Which property has a value? Quantity, units, basis and uncertainty convention.
Reference chain How is the result related to a reference? Named reference, qualification record and any limitations.
Lot and unit history Which physical material was used? Supplier lot, receiving record, storage history and use record.

Ask for evidence at the producer and material levels

ISO 17034:2016 addresses the competence and consistent operation of reference material producers. Its public scope includes both reference materials and certified reference materials. [4] For a purchasing review, record the producer, the claimed scope and the document applicable to the material. A general quality-management certificate answers a different question from a particular material’s value-assignment report.

For an internal working standard, request a qualification summary, the identity of the reference used, the approved intended use and the conditions that trigger reassessment. If the laboratory changes its measurement basis, it should review whether the existing qualification still supports the new report.

Build a reference-material enquiry that a laboratory can review

  • Name the measurement and the type of result required.
  • Identify the peptide form and any modification relevant to the comparison.
  • Request the assigned value, units, basis and uncertainty information when quantitative use is intended.
  • Ask for the material-specific certificate or qualification report, including its version.
  • Record lot linkage, storage conditions and the reference’s stated validity.

Use the existing guide to reading a peptide COA to review the offered lot. For a new project, connect the reference requirement with the analytical scope before comparing quotations. The research peptide catalogue identifies enquiry materials; any reference-standard role needs its own supporting record.

Frequently asked questions

Can a high-purity research peptide be used as a calibration standard?

It may be suitable after qualification for the intended measurement. A purity label alone does not establish an assigned quantitative value, its uncertainty or a reference chain.

What is the difference between lot traceability and metrological traceability?

Lot traceability follows the history of a material or document. Metrological traceability relates a measurement result to a reference through a documented calibration chain with uncertainty contributions.

Does every peptide reference need a certified mass-fraction value?

The required evidence depends on the measurement. Identity comparison and quantitative value assignment have different needs. State the task before choosing the reference material.

Which certificate should accompany a repeat reference-material purchase?

Request the certificate or qualification record applicable to the supplied lot and unit, including its current version, assigned properties and validity conditions. Keep it with the receiving record.
